The aim of the work is to isolate tyrosinase from Agaricus bisporus mushrooms, to determine its protein fractional composition, to study biochemical and physicochemical characteristics (protein, copper, monophenolase and diphenolase activities, pH and thermodependence of activity, thermal stability). Study of phenol oxidase activity inhibition of tyrosinase by benzylideneanilines and related compounds: determination of inhibition parameters (IC50, inhibition constant, type of inhibition); the influence of structural features, synergism with other inhibitors. Tyrosinase was isolated from Agaricus bisporus mushrooms by a modified method. It was found that the introduction of polyethylene glycol during the extraction process reduces the content of polyphenols by 3 times and increases the activity of the enzyme by 25%. The effect of 2-hydroxybenzylidene-2-aminophenol, 2,2-dihydroxyazobenzene, bis(2-hydroxyphenyl)-methane, 2,2-dihydroxydiphenyl ether, glyoxal bis(2-hydroxyaniline) and bisethylenediamine, 2,2-dihydroxybiphenyl on tyrosinase activity. It has been shown that the presence of the azomethine group in the molecule plays an important role in the inhibition of tyrosinase activity; 2-hydroxybenzylidene-2-aminophenol (IC50 6.5 µmol/dm3) exhibits the highest inhibitory ability. It was determined that when two inhibitors of tyrosinase activity were used together in various combinations, natural and synthetic compounds, inhibitors of the same or different mechanism of action, an additive effect was revealed. The simultaneous use of two enzyme inhibitors opens up great opportunities for the development of hyperpigmentation treatments, which helps to reduce toxicity due to a decrease concentration of both inhibitors.   Product Description popup.authors Illyushko Natalia О.
