Bogatsky National Academy of Sciences of Ukraine popup.description2  The objects of research are new derivatives of tryptanthrins, lysozyme, carbopol, gel. Synthesized tryptanthrine and its new derivatives (8-methoxy-indolo[2,1-b]quinazoline-6,12-dione, indolo[2,1-b]quinazoline-6,12-dione 6-oxime and 8-methoxy- indolo[2,1-b]quinazoline-6,12-dione 6-oxime), the chemical structure of the compounds was confirmed by the methods of mass spectrometry, IR and NMR spectroscopy. The interaction of tryptanthrins with lysozyme was studied, it was shown that in the range of their concentrations of 1.5-2.5 g/dm3, the hydrolytic activity of the enzyme remains at the initial level. By immobilizing lysozyme and including synthesized tryptanthrins in a hydrophilic matrix based on carbopol, a combined gel was developed for further evaluation of anti-inflammatory and antimicrobial properties of the compounds. The obtained gel is characterized by a high degree of spreading, which guarantees a good contact time of the formulation and maintenance of moisture in the place of application, and the stability of the triptanthrin suspension, which ensures the accuracy of the dosage of the drug. The hydrolytic activity of lysozyme remains at the level of 71%, immobilization occurs due to non-valent interactions with the carrier, which is confirmed by a decrease in the fluorescence intensity of the enzyme with an increase in the concentration of the polymer, as well as a change in the characteristic viscosity. The effect of immobilization with tryptanthrins on the operational stability of lysozyme was studied, it was shown that the pH-, thermal profiles of activity and stability of hydrolytic activity do not differ significantly from those of the free enzyme.
