Subject of the study - mRNA expression profile obtained as a result of total RNA sequencing, placental MSCs, umbilical cord endothelial cells. The aim of this work is to determine the angiogenic potential of individual subpopulations of placental mesenchymal stem cells. Methods - isolation and cultivation of human placental mesenchymal stem cells, isolation of placental mesenchymal stem cells clones, bioinformatic analysis of total transcriptome sequencing, flow cytometry, immunocytochemical study, directed differentiation, capillary formation in vitro. Results and their novelty. As a result of the study, clones of human placental MSCs were obtained and the whole genome transcriptome was studied by RNA sequencing. All clones, as during 9-10 passages of proliferation expressed at a low level of SC7, were heterogeneous in pan-cytokeratin (pCK) and alpha-smooth muscle actin (a-SMA). We also detected transcription factors characteristic of mesodermal cells, such as CDX2, EOMES, ERG. All studied clones had the ability to differentiate in osteogenic and adipogenic directions. It was found that clones similar to heterogeneous cultures of placental MSCs have the ability to form capillaries in vitro. Highly proliferative placental MSC clones are characterized by a relatively higher level of expression of genes involved in vascular development. The high level of pericytic gene expression may indicate the ontological relationship of MSCs with pericytic cells of the placenta. The SCC2 clone had the characteristics of stem cells with a high level of expression of genes involved in cell proliferation and autophagy.
