Registration Date 07-01-2023 Organization Institute of Cryobiology and Cryomedicine of the National Academy of Sciences of Ukraine popup.description2 Research objects: microalgae Dunaliella salina. The purpose of the work: to give a quantitative and qualitative assessment of the dynamics of β-carotene accumulation in D. salina cells depending on the composition of the medium and cultivation conditions, as well as to study the thermodynamic indicators of cooling-warming environments for the culture of this microalgae, which is an important stage in the development of effective methods of long-term low-temperature storage of biological objects. The work carried out and the analysis of the obtained data showed that the change in the composition of the culture medium (reduction in the content of microelements, increase in the concentration of sodium chloride) contributes to the directed synthesis of beta-carotene in the cells of the microalgae D. salina. A decrease in temperature to 4°С leads to a change in the pigment composition of cells after 24 hours - the synthesis of chlorophyll is inhibited and the synthesis of carotenoids increases. On the 30th day, the highest content of carotenoids is observed in cell cultures that were grown in conditions of a lack of trace elements or an increased content of sodium chloride. The main temperature range, within which damage to cells of D. salina microalgae probably occurs during cryopreservation, lies in the range from –10 to –25 °C, depending on the culture medium.
