Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2  According to the results of immunophenotypic analysis, about 45% of cases of acute myeloid leukemia (AML) were characterized by the presence of expression of cell surface marker CD34; at the same time, in 48% of cases, malignant cells was represented mainly by blast forms, and in 19% of cases, by cells in the late stages of differentiation. AML cases with a CD34+ phenotype of malignant cells were characterized by a higher level of DNA damage compared to CD34 ones, as indicated by a higher rate of double-strand breaks. In addition, in CD34+ cases of AML, the mRNA expression level of the c-MYC oncogene was also higher than the same indicators for CD34- AML cases and did not correlate with the degree of differentiation of these cells. Based on the results of the bioinformatics analysis, proteins that are involved in the processes of repairing DNA damage in cells and regulating the cell cycle were identified, and with which proteins CHK1, claspin and c-MYC proteins have a direct functional connection. The nature of mRNA expression of CHK1, claspin and c-MYC genes in tumor cell lines of myeloid origin was analyzed. In addition, an analysis of open databases was carried out and a list of the most mutated genes in AML and the nature of the most frequent chromosomal alterations for this nosology were identified.
