On the model of acute inflammation of the abdominal cavity in mice, the dynamics of the development of the therapeutic effect of both initial MSCs and MSCs that were preconditioned with hydrogen peroxide were studied. On the basis of these results, a method of assessing the therapeutic efficacy of human umbilical cord MSCs in vivo has been developed, which allows comparing different preparations of MSCs. Comparison of the effect of preconditioning of MSCs by various factors revealed the maximum increase in the therapeutic efficiency of MSCs under the influence of preconditioning with a low concentration of H2O2. This method of preparing MSCs for transplantation into rats on a model of acute pancreatitis led to an acceleration of the recovery of the pancreas and a real increase in the effectiveness of this process. The result of the project is establishing the possibility of manipulating the therapeutic effect of MSCs by preconditioning them during cultivation.
