Registration Date 28-01-2023 Organization Taras Shevchenko National University of Kyiv popup.description2 Objects of the research: molecular and genetic properties, diagnostic methods and epidemic potential of harmful isolates of plant viruses. The aim of the work: establishment of molecular, genetic and phylogenetic properties of dangerous viruses, optimization of methods of their molecular diagnostics and analysis of their epidemic potential in agro-ecosystems of Ukraine. Methods of the research: virological, serological, molecular biological and statistical methods. The phylogenetic properties of dangerous plant viruses were studied to understand the basis of their virulence and the ways of their evolution, the methods of molecular diagnostics were optimized and the epidemic potential of dangerous viruses in the agro-ecosystems of Ukraine was analyzed. It was established that the wheat dwarf virus isolates Khm-K-Ukr and MIP-12-Ukr belong to the phylogenetic clade E and, probably, have a common origin with the barley Polish isolate WDVLip and the wheat isolates from Ukraine WDV-Uk-Miron and Estonia. It was established that the highest rate of mutations in the CP gene is characteristic for the Khm-K-Ukr isolate from the Khmelnytsky region, which may indicate its increased epidemic potential and the risk of rapid spread and high harmfulness. Five cereal viruses circulate in Ukraine: WSMV, HPWMoV, BSMV, ВYDV-PAV і ВYDVMAV. WSMV isolates are characterized by high identity (>96.5%) with previously published wheat isolates from Ukraine and other European countries, and HPWMoV isolates are most similar (>85-98%) to isolates of this virus from the USA. In Ukraine, two viruses from the group of yellow dwarfism have been identified: ВYDV-PAV on wheat and BYDV-MAV on oats. The Ukrainian isolate BYDV-Ukr belongs to the species BYDV-MAV.
