The obtained results are useful for understanding the key intermolecular interactions that are essential for the initiation of fibrinolysis and the regulation of the rate of fibrin clot dissolution. It was shown for the first time that platelet microaggregates may play a role as the centers of initiation of fibrin formation from plasma fibrinogen. Activated platelets, via binding plasminogen and tissue plasminogen activator on their surface, act as centers of initiation of the clot lysis process. Approaches for the isolation and characterization of microparticles of platelet origin from donor’s blood plasma have been developed and optimized, and the presence of proteins of the fibrinolytic system in platelet extracellular microvesicles has been established. Qualitative and quantitative analysis of the proteomic composition of platelet microparticles can be used in the diagnosis of a wide range of diseases associated with platelet disorders. A highly sensitive and specific method for determining activity of PAI-1 (plasminogen activator inhibitor) was developed. The proposed method was tested for several pathological states (prothrombotic conditions, post-Covid syndrome), and recommendations for its use were developed. The molecular mechanisms of resistance of lung adenocarcinoma cells to apoptosis/anoikis induced by plasminogen/plasmin as an important condition for maintaining the migration of cancer cells and the formation of metastases have been studied. The obtained results have both scientific and practical value, while they expand the understanding of the mechanisms of cancer tumor metastasis, and therefore are valuable for the development of new effective approaches to pharmacotherapy of patients with an oncological profile.
