Registration Date 28-01-2023 Organization Institute of Food Biotechnology and Genomics of the National Academy of Sciences of Ukraine popup.description2  Using the electrophoretic analysis of gliadins, carriers of the null allele were identified among the offspring of F2 plants of soft winter wheat, heterozygous for the presence of the null allele of the Gli-B1 and Gli-D1 locus, and the separation of the Slaven soft winter wheat variety into biotypes by the Gli-D1 locus was carried out and a biotype with a null allele of the Gli-D1 locus was isolated. With the help of SDS-electrophoresis, genotypes were identified at the loci of high-molecular subunits of glutenins Glu-A1, Glu-B1, Glu-D1, and lines with a high potential score of baking quality (12) with a combination of alleles Glu-A1b, Glu-B1al, Glu-D1d were selected. Breeding and selection by habit of winter wheat lines with the absence of synthesis of omega-gliadins was carried out. Hybridization of lines with null alleles of Gli-B1 and Gli-D1 loci was carried out and F1 grain was obtained, which was sown on the experimental plot. Using microsatellite loci and the spike color locus Rg-B1, deletions in lines with null alleles at the gliadin loci Gli-B1 and Gli-D1 were mapped. The seed material of the lines from the crossing of Odeska chervonokola x B16 with null alleles at the gliadin loci Gli-B1 and Gli-D1 and with high-quality alleles of the Glu-1 loci was transferred to the Poltava State Agrarian University and the Plant Breeding Institute named after V.
