The purpose of the work: the development of biosensor portable analyzers for the quantitative determination of creatinine - a metabolite of protein metabolism, an important biomarker of the functional state of the kidneys - using as an analytical tool a recombinant form of stable creatinine deiminase (KDI) of microbial origin. A recombinant strain of Escherichia coli capable of overproducing the (His)6-tagged form of the enzyme from Corynebacterium glutamicum was constructed. The specific activity of the purified enzyme is up to 20 μmol per minute and mg of protein. Screening of nanocomposite mediators of electron transfer for chemosensing of ammonium ions was carried out and chemoselective membranes based on ammonium-sensitive Cu/Zn(Hg)S nanoparticles and based on electrodeposited polyaniline (PANi) in complex with Nafion were selected. Design and testing of a miniaturized microcurrent potentiostat compatible with modern gadgets has been carried out. Metrological studies of the portable creatinine analyzer were conducted and a set of scientific and technical documentation for the device was prepared. Laboratory regulations for the preparation of chemo- and biosensor elements of the analyzer have been prepared; programs and methods of technical testing of the device are optimized according to the requirements of the potential consumer; technical tests of an amperometric biosensor analyzer compatible with a tablet for the analysis of Crn content in human biological fluids were conducted. According to the results of the NDR in 2018 - 2022, 28 articles (with a total impact factor of 63.53), 5 abstracts of reports were published.
