A number of plasmids containing the RIB1, RIB6 and RIB7 genes (encoding the enzymes GTP cyclohydrolase II, 3,4-dihydroxy-2-butanone-4-phosphate synthase and riboflavin synthase, respectively) were constructed in various combinations and transformants of the wild-type and overproducers of riboflavin C. famata strains were obtained. It has been established that the expression of single genes, various combinations of two genes, and co-expression of all three genes leads to an increase in riboflavin production to a greater or lesser extent. The obtained results indicate that the coexpression of RIB1 and RIB6 genes is more important for the oversynthesis of riboflavin than the combination of other structural genes. A particularly high yield of riboflavin was found when cultivating the best of the engineered strains containing overexpressed genes RIB1, RIB7 SEF1 and RIB6 and having an improved supply of purine precursor, when grown on whey with the addition of ammonium sulfate. Thus, whey can be used as a promising substrate for the development of technology for the industrial production of vitamin B2. The obtained results made it possible to identify new promising targets for metabolic engineering of flavinogenic strains of C. famata in order to improve vitamin B2 production.
