Section 3. Development of reference methods for the arginine analysis with the usage of arginine-hydrolyzing recombinant enzymes. popup.stage_title Head Sybirnyi Andrii A., Registration Date 07-02-2023 Organization Institute of Cell Biology popup.description2 Scientific report: 71 p., 40 fig., 12 table., 68 sources. Objects of study: arginine-hydrolyzing enzymes – arginase (ARG), arginine deiminase (ADI), arginine oxidase (ArgO), functional electroactive nanomaterials, enzymatic methods for the determination of arginine (Arg). The purpose of the scientific research (Part No. 3, IBC of the NASU) is to develop reference methods for the analysis of Arg using Arg-hydrolyzing enzymes. The research was carried out during 2018-2022 in 5 stages. Research methods: biochemical, physico-chemical, analytical. As a result of the work, new reference methods for determining the content of Arg using arginine-hydrolyzing enzymes were created: an enzymatic-chemical method based on ARG and two types of monoenzyme amperometric biosensors (ABS). ABS contained enzymes co-immobilized on synthesized electroactive nanoparticles (NPs): 1) ADI on ammonium-selective NPs, 2) ArgO on NPs with pseudo-peroxidase activity. The suitability of new methods for determining the content of Arg in real juice samples has been demonstrated, a close correlation of the data of ABS analysis with the results obtained by enzymatic-chemical and traditional reference methods has been established. According to the results of the research during the reporting period, 14 articles were published (with a total impact factor of 45.65), 2 chapters in monographs and 19 conference proceedings. The work was completed in full in accordance with the plan. Keywords: ARGININE, ARGINASE, ARGININE DEIMINASE, ARGININE OXIDASE, ENZYMATIC METHODS, FUNCTIONAL ELECTROACTIVE NANOMATERIALS, BIOSENSORS.
