Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2 It was established for the first time that spermine (Spn) shows pronounced growth inhibitory effect on the human prostate cancer cell line LNCaP. The vitality was increased by the combined action of Spn and aminoguanidine. Spn at a concentration of 1.5 – 5 mM had impact on emerging of cells with signs of apoptosis. Similar effect of 5 mM Spn and 15 mM etoposide on the proportion of LNCaP cells in early and late stage of apoptosis was shown. Injection of Spn led to changes in the architectonics of cells. Growth inhibitory effect of the Spn was accompanied by disorders of the cytoskeleton, adhesive properties and intercellular contacts, membrane disintegration, absence of pseudopodia, emersion of cells with hyperchromatic nuclei and double nuclei and micronuclei. In the cell preparations with Spn at a dose of 5 mM, the complete absence of mitosis was noted. Growth inhibition was accompanied by a decrease in the expression of ODC, c-myc genes and in the levels of putrescine and spermidine in cells, an increase in Spn and acetylspermidine, decrease in the cell proliferation index – the ratio of spermidine/spermine. The Spn affected in changes of cell distribution according to the ζ-potential values, which indicated an increase of their adhesive properties and their transition to apoptosis. Obtained data is a basis for the development of new effective regimens for antitumor therapy of prostate cancer based on the use of spermine and inhibitors of its catabolism.
