Therefore, it is important to determine the molecular mechanisms that underlie the occurrence of childhood tumors with the aim to develop the personalized treatment. It is known that the cellular RB-E2F1 pathway is inactivated in such tumors. Therefore, the search for proteins that control the abovementioned and other cellular pathways is an important and urgent task of modern oncology. A promising candidate is the MRPS18-2 protein, that interacts with RB and induces primary cell transformation. To achieve our goal, i.e., to find out a role of the MRPS18-2-RB interaction in the control on cell stemness and in development of childhood tumors, the expression pattern of the MRPS18-1-3 and RB1 genes was studied. Gene mRNA levels were assessed by qPCR in clinical samples (blood serum and tumor tissue); protein signals were determined in childhood tumors - glioma and medulloblastoma - by immunohistochemical and western blot analyses. The MRPS18-2 and -3 genes are expressed in tumor tissues at higher levels compared to controls. In addition, the MRPS18-2 protein signal is dramatically increased in medulloblastoma glioma cells compared with healthy brain tissue.
