Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2  On the basis of the bioinformatics analysis carried out using the miRDB, TarBase v.8.0 and MicroT-CDS databases, a miRNA, involved in the regulation of the expression of extracellular matrix genes (MMP1, MMP8, SPP1 and SPARC) were identified, namely hsa-mir-29b-2-5p , hsa-mir-146a-5p, hsa-mir-181a-5p, hsa-mir-221-5p. It was found that the main signaling pathways that regulate the identified miRNAs and play an important role in the onset and progression of PCa are the apoptosis signaling pathway, the inflammatory signaling pathway mediated by chemokines and cytokines, and the integrin signaling pathway. The study of expression pattern of extracellular matrix genes at the mRNA levels established that the mRNA levels of SPP1 and SPARC in the tissue of PCa were 2.9 times and 77.6 times higher in comparison with the tissue of BPH. It was determined that the tissue of patients with BPH was characterized by significantly lower mRNA levels of type I and II collagenases compared to the tissue of patients with PCa. Immunohistochemical study of the expression of extracellular matrix proteins demonstrated a significantly higher level (p < 0.05) of osteonectin and MMP-8 in the tissue of the PCa, respectively, compared to the tissue of the BPH. It was found that the levels of hsa-mir-146a-5p and hsa-mir-221-5p were 1.6 and 1.2 times lower in the tissue of BPH compared to similar indicators in patients with PCa. The obtained data testify to the expediency of researching the expression pattern of vegeto-associated collagenases for the differential diagnosis of PCa.
