Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2 It was proved that the transition of Lewis lung carcinoma (LLC) cells from a glucose-deficient environment to deadhesive growth in deficit-free environment determined (in comparison with the corresponding indicators during adhesive growth) a statistically reliable: decrease in the number of living cells by 33%; a decrease in the percentage of cells in the G1 phase of the cell cycle by 12% and an almost two-fold increase in the percentage of cells in the G2 phase; a threefold increase in the percentage of apoptotic cells and an increase in the intensity of reactive oxygen species production; an increase in the rate of glucose consumption by 50% and lactate production by 70%; a two-fold decrease in the activity of lactate dehydrogenase (LDH). Using the LDH inhibitor, it was proved that regardless of metastatic potential, Lewis lung carcinoma cells are heterogeneous with regard to the involvement of aerobic glycolysis in their growth and survival processes. Approximately one-third of cells from both highly metastatic Lewis lung carcinoma (LLC) and low metastatic (LLC/R9) lines were able to survive and proliferate when aerobic glycolysis was inhibited by oxamate. Oxamate-resistant subpopulations of tumor cells of both lines practically do not consume glucose, but retain a fairly high proliferative activity, which is not provided by glycolysis, but most likely due to a non-canonical metabolism of glutamine.
