When conducting research on experimental animals of various ages, healthy people and patients with metabolic disorders on the basis of ICAMER in Elbrus, at the Institute of Physiology named after O.O. Bogomolets of the National Academy of Sciences of Ukraine and clinics in Kyiv characterized the role of microRNA- dependent regulatory factors in cellular and metabolic adaptation in various hypoxia regimes, aging and metabolic disorders, determined the cellular and molecular mechanisms of the development of metabolic disorders, in particular, insulin resistance and type 2 diabetes. Using the effects of acute, chronic, periodic and combined hypoxia regimes in highland and plain conditions and in experimental metabolic disorders, changes in the expression of microRNA-1 and -34a, metabolic regulatory proteins IGF-1, p53, apelin, leptin, pAMPK in rat tissues, the mechanisms of metabolic compensation and adaptation to oxygen deficiency mediated by them and their impairment during aging. In patients with metabolic syndrome and type 2 diabetes, a violation of microRNA-1- dependent regulation of insulin-independent compensation mechanisms, in particular, hypoxic induction of IGF-1, was found. The favorable effect of the developed hypoxic correction regimes on the indicators of carbohydrate and lipid metabolism was established. The effect of periodic hypoxia also led to the intensification of microRNA-1/IGF-1 and microRNA-34a/p53-mediated regulatory mechanisms, improved insulin-independent mechanisms of compensating carbohydrate metabolism disorders. The obtained results make it possible to create approaches to genetic-based methods of prevention and correction of age-related diseases, including cardiometabolic ones.
