In the course of the work, nine different freezing protocols were tested. The programs differed from each other in terms of freezing rate, sample cooling period, and final freezing temperature. The object of the study was human mesenchymal stem cells. The viability of MSCs was compared after the procedure of freezing and short-term storage at -1600C, in vapors of liquid nitrogen, using flow cytofluorometry, according to the intensity of inclusion of 7AAD dye. As a result of the work, it was demonstrated that for the suspension of individual cells, the cooling period of the sample, the cooling rate has a significant effect on the further viability of the cells. Thus, shortening the cooling period, with a fairly high freezing rate, ensured high cell survival. Conversely, lengthening the cooling period with slow freezing resulted in a high percentage of dead cells after thawing. The obtained data will make it possible to adjust the freezing protocols of individual cells, cell suspensions, and human tissues. Object of research: mesenchymal stem cells. The purpose of the study was to compare the effectiveness of freezing programs on the viability of MSCs and to reveal any specific dependence of cells on the conditions of adaptation of preservation and storage at very low temperatures. The objectives of the study were to investigate the viability of MSCs after thawing and long-term storage, which had previously been initiated by specific low-temperature freezing using a software freezer. As a result, it was found that the viability of the cell suspension is significantly affected by the duration and speed of cooling, the speed of freezing, and the depth of the entire freezing. Scientific and practical significance. This stage of research provides clarification and understanding of the preparation of mesenchymal stem cells for long-term preservation at very low temperatures.
