Subject of the research – biosensors and enzymatic approaches for determination of creatinine. The purpose of the work - the development of new bioanalytical methods for quantitative determination of creatinine - an important biomarker of the functional state of the kidneys. Research methods: chemical, physicochemical, biochemical, microbiological, enzymatic, electron microscopy, X-ray spectral microanalysis, UV-visible spectroscopy, nuclear magnetic resonance spectroscopy, Raman spectroscopy, potentiometric method, enzyme immobilization methods, statistical analysis. Results and novelty: For the first time: – the codaA gene of Corynobacterium glutamicum was cloned in the cells of the parental strain and the activity of CDI in the obtained transformants was investigated; - an enzymatic method of creatinine analysis was created for detection of N-methylhydantoin, the final product of creatinine hydrolysis; - transition metal nanoparticles capable of forming a redox-active complex with N-methylhydantoin were synthesized; - an amperometric chemosensor was developed for the analysis of N-methylhydantoin using nCoCu nanoparticles; - pH-sensitivity, stability of transducers based on pH-sensitive field-effect transistors and their functionality were investigated. Improved: – the method of one-step purification of creatinine deiminase by the use of metal affinity chromatography; – the methodology for obtaining NPs of noble, transition and rare metals, as well as their hybrid forms by methods of chemical reduction of the corresponding salts; – the method of immobilization of creatinine deiminase on the surface of electrochemical transducers.
