Purpose of work: to determine the common and different mechanisms of geroprotective action of mitochondrial respiration effectors (2,4-dinitrophenol (DNP) and alpha-ketoglutarate (AKG), certain phenylpropanoids (ferulic acid) and plant preparations with a high content of phenylpropanoids (anise hyssop, chili pepper and Rhodiola rosea) in model animals, fruit fly Drosophila melanogaster and mice. Research methods – biochemical (determination of functional activity of mitochondria, oxidative stress markers, activities of antioxidant and glycolytic enzymes, levels of storage and intermediate metabolites); physiological methods (breeding of fruit flies and mice, assays of lifespan and functional aging in fruit flies, behavior reactions and food consumption in mice), molecular biology methods (ELISA, western-blot, PCR), histology methods (classical and cryohistology), methods of mathematical statistics. Results and novelty: It has been shown for the first time, that AKG and ferulic acid are able to correct metabolic changes caused by a high-calorie diet high in animal fats and fructose and a cafeteria diet. The protective effect of AKG was manifested in the reduction of anxiety-like behavior in mice, diminishing of oxidative stress and inflammation with an increase in the power of antioxidant defense and normalization of energy metabolism in mouse tissues. The addition of ferulic acid to the cafeteria diet did not correct body weight gain and visceral fat accumulation, but normalized blood parameters, improved liver function, which was accompanied by a decrease in the expression of genes related to autophagy, antioxidant defense, and regulation of energy metabolism.
