The aim of research is to investigate the proliferative and metabolic activity of human mesenchymal stem cells (MSCs) in monolayer and three-dimensional media; to optimize a non-healing wound model to determine the therapeutic potential of these cells. Optimal conditions for obtaining multicellular homogeneous spheroids were established. It was shown that MSCs during 3D cultivation in the composition of spheroids, hydrogels based on alginate and blood plasma, as well as macroporous scaffolds based on blood plasma maintained high viability and potential for multilineage differentiation, but had significant differences in the ability to spread and proliferate which were accompanied by the restructuring of the MSC cytoskeleton, which depended on the type of 3D structure. Taking into account the obtained results, it was established that the plasma-based hydrogel is a suitable cell carrier for wound treatment, therefore it was used to study the effect of MSCs cultured in a monolayer on the healing of complicated wounds in mice. Application of MSCs in a plasma gel carrier to wounds treated with 0.5 mg/ml mitomycin C promoted earlier maturation of granulation tissue compared to wounds that healed spontaneously. At the same time, it was shown that the used concentration of the healing inhibitor did not allow to fully evaluate the therapeutic potential of MSCs, therefore, the dependence of the healing rate on the concentration of mitomycin C was studied and its optimal concentration was chosen – 2 mg/ml, which will be used for further experiments.
