The purpose of the work is to study the possibilities of using various protein/non-protein drug delivery systems (functionalized collagen matrix, alginate capsules, EMAR II cytokine nanocomposite complexes, bacterial cellulose composites and amniotic fluid exosomes) to increase their therapeutic effectiveness. Studies of alginate capsules have shown that they can be used for effective delivery of therapeutic proteins. It was shown that MSCs encapsulated in alginate capsules and transplanted intraperitoneally to rats with liver cirrhosis lead to faster liver recovery compared to systemically administered MSC suspension. Approaches to the stabilization of the EMAP II structure were developed. The analysis of spatial structures revealed that one of the key roles in the formation of contact between protein molecules in oligomers is played by the unstructured loop 34DVGEIAPR41, which blocks the hydrophobic tryptophan "pocket" on the surface of the protein. The interaction between the active center and the unstructured loop is hydrophobic and electrostatic in nature. According to computer modeling of the structure of EMAP II complex with dextran-70, individual dextran molecules block potential sites on the surface of the protein responsible for the formation of EMAP II aggregates, which is confirmed by experimental data on stabilization of the complex obtained by dynamic light scattering. During the study of composites of bacterial cellulose and amniotic fluid exosomes, amniotic fluid exosomes were isolated, their properties were studied, and they were introduced into a bacterial cellulose film. Exosomes in the composition of the composite activated the migration and proliferation of mouse fibroblasts in contrast to the control sample based on sterile pbs buffer and cellulose film.
