The optimal method of enzyme immobilization was chosen based on the activity and stability of enzymes and their functioning parameters in the immobilized state. Based on the constructed mathematical model, the optimal conditions for the immobilization of lactate oxidase and pyruvate oxidase as part of the bioselective elements of monobiosensors were selected. In addition, the influence of the characteristics of the solution on the functioning of these monobiosensors was analyzed. The effect of pH, buffer capacity, ionic strength of the solution and other parameters on the operation of monobiosensors was investigated. The obtained results should be taken into account when using these biosensors when working with real biological fluids. The reproducibility of biosensors during continuous operation, operational stability and stability under different storage conditions were studied. The selectivity of both monobiosensors with respect to a number of possible interferents was also analyzed and ways to improve this parameter by using additional semipermeable membranes based on polymetaphenylenediamine were proposed. The proposed monobiosensors were combined into a single biosensor system for simultaneous determination of pyruvate, lactate concentration and lactate/pyruvate ratio. The simultaneous functioning of all sensor elements in one system has been tested and optimized. Cross influence of substrates (lactate, pyruvate) and cofactors on the functionality of both enzyme monobiosensors as part of the biosensor system was studied.
