The subject of research: molecular mechanisms of BCR-ABL functioning, the role of Bcr domains in the functioning of normal cells and cells transformed by the BCR-ABL chimeric oncogene, the role of transcription factors in blast crisis, the development of drug resistance in prostate tumors. Methods: Standard molecular biological protocols, which included: isolation of RNA and DNA, oligonucleotide primers design, reverse transcriptase polymerase chain reaction (RT-PCR), PCR, real-time PCR, creation of recombinant DNA, Southern hybridization, DNA sequencing, fluorescence confocal microscopy of live and fixed mammalian cells, ultra-high-resolution fluorescence microscopy STED, etc. Work results. Under the action of imatinib, as inhibitor of the kinase activity of the oncoprotein, a some of the signaling pathways are disrupted, which makes it impossible to export the Bcr-Abl oncoprotein to the cell nucleus. Also, under the condition of imatinib, the expression of the ZNF217 protein stops, which can be an important prerequisite for reducing the proliferative activity of cells and the beginning of their apoptosis. The formation of ternary complexes between cortactin, actin and BCR was shown by the method of ultra-high-resolution confocal and STED microscopy on K562 culture.
