PCR analysis of individual grains from ears of F2, F3 and selection of genotypes according to the target gene was carried out. Sowing of families represented by F3, F4 grains with carriers of the target gene was carried out, a review was carried out by sowing. PCR analysis of grains from ears of F4-F5 from crosses with carriers of Sr genes was carried out; testing of parental forms for the presence of target Sr genes and determination of their alleles of prolamin loci by electrophoresis of grain proteins. For the use of gene-specific primers Sr33A, winter wheat genotypes with the Sr33 gene (homozygotes and heterozygotes) were selected in the population of F2 and F3 ears. Carriers of the Sr33 gene were identified among the population of F4 and F5 ears. Alleles of the stem rust resistance genes Sr33 were confirmed in lines DK20, DH31, DH-G76-54, and Sr39 and Sr40 in RL6089 and RL5711, respectively.
