Genetic studies of patients with primary DYT2 dystonia have identified mutations in the gene for the neuronal calcium-sensing protein, hypocalcin (HPCA), as the cause of the disease. However, how these mutations lead to movement disorders is still unknown. The brain of patients with primary dystonia does not contain obvious pathological abnormalities, while functional studies indicate its insufficient inhibition and abnormal plasticity. We recently found that inhibitory control in neurons is significantly impaired upon expression of the HPCA mutant DYT2. It is known that the HPCA can function as a sensor in long-term depression, a widespread form of synaptic plasticity. Using a multidisciplinary approach, we plan to establish how HPCA regulates long-term depression and whether expression of its DYT2 mutants leads to a reduction of this depression. We expect that long-term depression may be disrupted by DYT2 mutation leading to significant excitation of neural networks and motor disorders, and expression of wild-type HPCA may be a therapeutic strategy for effective treatment of DYT2 dystonia.  Product Description popup.authors Dovgan Olexandr V.
