The goal of the project is to apply bioinformatics methods for normalization and integration of data groups in different gestational periods, overcoming the problem of small samples; quantitatively characterize how the expression of genes in the placenta tissue changes between the trimesters of pregnancy in normal and PE; and in which cells, using mRNA sequencing data in isolated placental cells, and also where differential expression takes place in 3-dimensional tissue space by in situ hybridization. Generalized, quantitatively characterized and updated data on gene expression in the human placenta according to cellular specificity and spatial localization of expression, normal gestation periods and PE will deepen knowledge about the pathogenesis placenta development in PE. An integrative analysis of gene expression in the human placenta between the first and second trimester of pregnancy was carried out. 251 differentially expressed genes were identified. According to the clustering data, the most represented genes were found, which are responsible for immune processes and for angiogenesis and morphogenesis. Among the genes responsible for immune processes, genes were found that recognize harmful factors, neutralize or inactivate them. Genes that contribute to the recruitment of immune cells that contribute to the pro-inflammatory response and genes that support the mother's tolerance to the semi-allogeneic fetus have been identified. This project is the first part of an ambitious project related to the search for markers of pregnancy complications caused by placental dysfunction.
