The purpose of the work is to determine the background frequency of damage to the genome of human somatic cells by cytogenetic and molecular genetic indicators. Research methods are molecular genetic (single-cell gel electrophoresis method - Comet assay), cytogenetic analysis of chromosomal aberrations, analytical, statistical. During the work, a comprehensive approach to the determination of radiation-induced (target) and radiation-inducible (non-target) damage to the genome was tested from the standpoint of the combined use of cytogenetic, molecular genetic and epigenetic research methods. As a result of the study, the frequency and spectrum of chromosomal aberrations, the number of aberrant metaphases (1.64 ± 0.34) %, the degree of damage to the DNA molecule by the level of single- and double-strand breaks (6.34 ± 0.72) %, the intensity of apoptosis ( 0.91 ± 0.33) % and methylation level in peripheral blood lymphocytes of conditionally healthy volunteers. The obtained background indicators, which characterize the state of the genome of somatic cells of conditionally healthy individuals, correspond to the limit norms. The obtained results will be used to improve the diagnostic criteria for predicting the course of the disease and to choose an adequate treatment strategy for patients with a history of radiation.
