LM Litvinenko of the National Academy of Sciences of Ukraine popup.description2  As a result of this research, a general approach to the synthesis of 2-mercapto-1,3-thiazolopyridines and 2-mercaptoimidazopyridines based on heterocyclization of o-halogenaminopyridines derivatives with sodium xanthate or carbon disulfide in pyridine in the presence of organic bases – triethylamine was proposed. The synthetic limits of these reactions, depending on the reagents and reaction conditions, have been studied, which makes it possible to vary widely the substituents in the pyridine cycle. A method of obtaining variously substituted 2-chloro-1,3-thiazolopyridines by the action of chlorine on 2-mercapto-1,3-thiazolopyridines in the hydrochloric acid was developed. The method gives high yields of 2-chloro-1,3- thiazolopyridines and high product purity (90%). A method for obtaining polysubstituted methyl 2-R1,R2-amino-1,3-thiazolopyridines by nucleophilic substitution of chlorine in position 2 on the amino function was developed. Carbonylation of 2-chloroderivatives of thiazolopyridines was studied. The possibility of selective synthesis of carboxymethyl derivatives of thiazolopyridines is shown. For the first time, methyl imidazo[4,5-b]pyridine-6-carboxylates were synthesized by carbonylation of 6-bromimidazopyridine derivatives. A large array of 1,3-thiazolopyridines containing various substituents in all positions of 1,3-thiazolopyridines, which can be used to create new medicinal products, has been synthesized.
