Tthe investigation of structural state modifications and the development of oxidative processes in nucleated cells of human cord blood, including stem hematopoietic cells, and erythrocytes under the influence of various cryoprotective solutions and antioxidants, as well as the study of the pathways of the development of apoptosis and the possibility of its prevention to enhance cell stability during various stages of cryopreservation and after transfer to physiological conditions in vitro. Research methods: cryobiological, biochemical, biophysical (light and fluorescence microscopy) methods, the method of ductal cytofluorimetry, the method of electrophoresis in Ds-Na-PAGE gel. Utilizing modern research methods, it was established that the addition of antioxidants such as glutathione in a final concentration of 1-3 mM, trolox (50-70 μM) or L-carnitine (15-20 mM) to DMSO-containing cryoprotective solutions for cryopreservation of nuclei, including hematopoietic progenitor, human cord blood cells, leads to a reduction in the number of cells with excess ROS content, which allows for the preservation of a larger number of viable cells after thawing. It is demonstrated that the use of these antioxidants enables a reduction in DMSO concentration from 7.5-10% to 5% without significant loss of the number of preserved and viable cells. The the potential to enhance ROS formation in erythrocytes under the influence of cryoprotectants and correct the process with the aid of antioxidants is shown. The fundamentally new experimental data obtained provide a solid foundation for substantiating and developing new methodological approaches to enhance the preservation and long-term survival of human donor and cord blood cells after thawing for the uninterrupted supply of military hospitals and other medical institutions with high-quality blood components and preparations during martial law and peacetime.
