Conducted experimental studies using a complex of methods (clinical, surgical, cell culture, immunological, morphological, light, fluorescent, polarization and electron microscopy) revealed a more pronounced activation of the structures of the chondrogenic and fibrous layers of the cartilage plate at the border of the defect after local and intravenous injection of MSCs. An increase in the functional activity of chondroblasts, chondrocytes and fibroblasts with the manifestation of paracrine interaction with the participation of vesicular formations during the local application of allogeneic MSCs in the area of the cartilage defect was revealed. The activation of the restoration of cartilage matrix structures 1, 2 months after the operation and wound healing with the formation of fibro-chondroid tissue and the subsequent maturation of the scar after 3 months were registered. Activation of wound healing with systemic and local administration of MSCs with the participation of immune processes and changes in the dynamics of pro-inflammatory (IL-6) and regenerative (TGF-β1) cytokines in blood serum was shown. No significant difference in the development of reparative processes was found after the use of native and preconditioned cells and restoration of the typical structure of cartilage tissue. The use of allogeneic MSCs to increase the efficiency of reparative processes of cartilage tissue is promising and allows recommending the use of local and intravenous MSC administration. The use of local and intravenous administration of allogeneic MSCs is promising for increasing the efficiency of reparative processes of cartilage tissue and allows recommending their implementation.
