M.V.Zubets of National Academy of Agrarian Science of Ukraine popup.description2 It was established that a 5% solution of polyvinyl alcohol (PVA) is the most optimal for the immobilization of boar spermatozoa for the purpose of sperm injection into the egg cytoplasm (ICSI), compared to 10%, 1% and 0.1% solutions. It is shown that it is impractical to use the commercial Sperm Preparation Medium (Origio, Denmark) for artificial insemination preparation of deconserved ejaculated boar spermatozoa. It was established that the optimal medium for the selection of spermatozoa by the flotation method is Sp-TALP (Parrish J.J. et al., 1988), in which 69.7±3.1 million/ml spermatozoa were obtained, of which 6.8±0.56 million/ml were mobile. Additional methods of assessing the quality of boar spermatozoa (the level of DNA fragmentation) have been applied to increase the efficiency of obtaining embryos in vitro, which is necessary for the expansion of research on the preservation of domestic pig breeds. It was established that the average level of DNA fragmentation of cryopreserved spermatozoa of boars of the Myrhorod breed is 84.2% (boar Dnipro 641 - 82.0%, Kamysh 853 - 86.3%), wild boars of the Ukrainian steppe white breed - 96.5% (boar Aspect 2029 – 97.3%, Arsenal 1669 – 95.7%).
