Registration Date 30-01-2024 Organization Institute of Cell Biology and Genetic Engineering of the National Academy of Sciences of Ukraine popup.description2  The goal of the second stage of the project was to determine the regularities of the functioning of the genetic system of stomata biogenesis and to identify potential natural sources of valuable alleles of genes that determine the number and morphology of stomata, for further manipulation of leaf surface structures using biotechnological approaches to increase wheat drought resistance. The following set of works was carried out: the peculiarities of stomatal morphology were investigated and their quantitative parameters were evaluated according to stomatal density, length and width of closing cells, length and width of stomatal gap for 10 varieties of wheat; the expression profiles of the homeologous genes FAMA, SCR, SHR and FLP were analyzed; rational conditions for the amplification of DNA markers designed to detect polymorphisms of promoter genetic sequences EPF1, EPF2 and MUTE were selected; screening of 3 collections of wheat varieties and lines was carried out in order to find valuable alleles of stomatal biogenesis genes using developed DNA marker systems; constructed plasmids for point editing of EPF1 and EPF2 genes, integrated these plasmids into E.coli, dH5a strain, confirmed the integration of plasmids into the bacterial genome by selecting single colonies on a selective medium and PCR analysis for the presence of the CAS gene, carried out approbation and optimization of the agrobacterial technique transformations with the model construct pCB271 (GFP gene); a comprehensive comparison of the proteomic profiles of the drought-tolerant Odeska 267 winter wheat variety and the drought-sensitive Darunok Podillya variety under the temporary effect of drought at the flowering stage was performed; drought resistance markers are proposed.
