Registration Date 05-02-2024 Organization Odessa I. I. Mechnikov National University popup.description2  Strains of marine bacteria were isolated from the deep-sea sediments of the Black Sea, their biological characteristics were studied, and identification was carried out based on the spectra of fatty acids and the results of 16S rRNA sequencing. A variety of biologically active compounds with antitumor activity have been established using metabolomics methods. The 10 most active strains of marine bacteria - producers of antitumor compounds - were selected from 210 investigated strains in tests for intercalary and cytotoxic activity on cell cultures of human rhabdomyosarcoma and adenocarcinoma of the larynx, cholangiocarcinoma and hepatocellular carcinoma of mice. Cytotoxic activity was established for the first time in marine sporulating bacteria of the genus Sutcliffiella. The combination of marine actinobacteria extracts with a standard chemotherapeutic drug (gemcitabine) demonstrated pronounced antitumor efficacy against murine cholangiocarcinoma (iCCA) cells. The method of flow cytometry showed that the mechanism of the antitumor effect of extracts of marine actinobacteria on the culture of cholangiocarcinoma of mice is related to the induction of early apoptosis and cell senescence. The growth patterns of the producer strain S.ambofaciens 561 and the optimal conditions for its cultivation in the New Brunswick Scientific BioFlo®310 (USA) fermenter for the accumulation of metabolites were established. Recommendations have been developed for further studies of selected strains of producers of active antitumor compounds on laboratory animals for induced carcinogenesis in vivo for a more detailed assessment of the effectiveness of combined therapy Product Description popup.authors Volodymyr O.
