A method of CIK viability monitoring was developed by assessing their metabolic activity due to glucose consumption. The obtained cellular product was characterized by a high content of functionally active CD3+CD56+killer lymphocytes (cytotoxic activity) against tumor cells. Lectin B. subtilis IMB B-7724 doesn't cause a toxic effect on human T-lymphocytes and can act as a mitogen. It was first discovered that lectin in human ВС cells activates the apoptotic program in a Caspase 3/7-dependent way. The cytotoxic effect of CIК against MCF-7 and MDA-MB-231 BC cells can be enhanced by incubation with lectin B. subtilis IMВ B-7724. In a 3D model of BC cell culture, it was proven that lectin does not affect the size of spheroids, but prevents the spread of tumor cells. The obtained data show the promise of the combined use of bacterial lectins and cytokine-induced killer T-cells as a new strategy for cancer biotherapy.
