НАНУ Registration Date 19-02-2024 Organization Institute of Cell Biology of the National Academy of Sciences of Ukraine popup.description2  Recombinant strains with increased expression of ZWF1, SOL3 and GND1 genes, which encode enzymes of the oxidizing step of the pentose phosphate pathway, were constructed. The effect of increased supply of riboflavin precursor - ribulose-5-phosphate on the oversynthesis of this vitamin by engineered strains was investigated. It was established that overexpression of GND1 gene (coding for 6-phosphogluconate dehydrogenase) increases riboflavin production in Candida famata L20105 by two times and by 1.3-1.5 times in riboflavin overproducers BRP and AF-4. The use of lactose as a carbon source leads to an increase in the synthesis of riboflavin by these strains by 1.4-2 times; Overexpression of SOL3 gene, which encode 6-phosphogluconolactonase, leads to a 1.3-fold increase in riboflavin production compared to the recipient strain AF 4. In other strains L20105 and BRP, overexpression of this gene did not lead to an increase in the level of riboflavin synthesis. Overexpression of the ZWF1 gene (encoding glucose-6-phosphate dehydrogenase), on the other hand, suppresses the growth and synthesis of vitamin B2 by the yeast C. famata. The revealed regularities of the regulation of riboflavin synthesis can be used for the construction of new stable superproducers by combining in changing the expression of several regulatory factors.
