The purpose of the work is to carry out barcoding, clarify the taxonomic status and assess the genetic diversity of rare species of the flora of Ukraine. Molecular genetic (PCR, molecular markers, cloning of DNA fragments, sequencing) and bioinformatics (analysis of DNA sequences, construction of philodendrograms, work with databases) methods were used. Effective monitoring of flora, assessment of viability of existing populations and rational management of biological resources are impossible without accurate identification of species and assessment of genetic diversity at the inter- and intra-species levels. In the work, selected regions of the chloroplast (matK, trnT-L, trnL-F, trnS-G, trnH-psbA) and the nuclear (ITS1-2 and 5S rDNA) genomes were sequnced, bioinformatics databases were searched, species affiliation was determined using molecular barcoding, the taxonomic status was clarifiedand and the genetic diversity was assessed for 45 rare species of the flora of Ukraine, which belong to the genera Aconitum, Lathyrus, Limonium/Goniolimon, Muscari and Tulipa. SRAP markers were used to assess intraspecific (population) genetic variability. The genetic uniqueness of Ukrainian representatives of the complex species A. anthora s. l. and T. sylvestris s. l. was demonstrated. It is shown that the Ukrainian samples of M. botryoides belong to a new, previously undescribed species. The hypothesis about the suspected hybridization between L. vernus and L. venetus species, which can lead to the disappearance of L. venetus in natural populations was confirmed.
