Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2 We collect tumor samples and peripheral blood of patients with embryonic tumors, as well as primary cultures of embryonic tumors and established cell lines. 32 samples of medulloblastoma, 37 samples of glioma and 15 samples of chordoma were collected. The expression pattern of the MRPS18 family genes at the mRNA and protein levels was assessed in a proportion of ex vivo samples. Sub-lines of human tumor cell lines were created, namely Kelly-MRPS18-1-3, WERI-MRPS18-2, WERI-MRPS18-1, WERI-MRPS18-2-RB and A375-MRPS18-1-3. Using the cocktails of chemicals, the WERI-MRPS18-2 and WERI-MRPS18-2+RB retinoblastoma sub-lines were differentiated into osteoblasts, as shown by an Alizarin red qualitative reaction, and into chondrocytes, confirmed by an Alcian blue qualitative reaction. We have previously shown that primary fibroblasts where MRPS18-2 was overexpressed, acquired the properties of stem cells and differentiate into osteoblasts, chondrocytes, and adipocytes in vitro. Thus, a role of the MRPS18-2 protein in controlling the functional plasticity of cells is universal, making this protein a target for anticancer therapy.
