Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2 It was established that MDL-72527 and chlorhexidine (inhibitors of spermine oxidase, SMO) significantly enhance the cytotoxic effect of spermine on hormone-sensitive (LNCaP) and hormone-resistant (DU-145) human prostate cancer cell lines. Their combined use leads to inhibition of cell growth at much lower concentrations than when these agents are used separately. During the implementation of the project, molecular biological and biochemical indicators were verified, the changes of which correlate with apoptotic changes and cell viability, and which are appropriate to use to evaluate the restinhibitory effect of spermine and SMO inhibitors, namely: a decrease in the expression levels at the protein level of the genes ODC1, AZIN, c -Myc, Bcl-XL, an increase in the level of spermine, a decrease in the value of the spermidine/spermine molar ratio, and an inversion (change of sign from negative to positive) of the total surface charge of cells.
