It has been shown that the protective mechanisms of hydrogen sulfide and glutathione action are associated with suppression of oxidative stress, restoration of constitutive Ca2+-dependent NO synthesis, activation of ATP-dependent K+ channels and effect on intracellular structures, namely non-specific megachannels as mitochondrial transient conductance pores, which are one of the key regulators of the functional state of mitochondria. Hydrogen sulfide and glutathione have been shown to be protective in cardiac ischemia-reperfusion injury and to play an important role in restoring endothelium-dependent vascular relaxation. We have demonstrated ways to restore the insufficient synthesis of endogenous glutathione and H2S using various modulators, namely pyridoxal-5-phosphate, a combination of propargylglycine and the amino acid L-cysteine. Their use has a protective or corrective effect on cardiac dysfunction in certain conditions caused by insufficient synthesis of these natural antioxidants. Thus, stimulation of endogenous glutathione synthesis significantly increased functional reserves and myocardial resistance to ischemia due to inhibition of ROS formation in cardiac tissue and preservation of mitochondrial function. Thus, the cardioprotective and vasculoprotective properties of hydrogen sulfide and glutathione were mainly due to their ability to suppress oxidative stress and the development of mitochondrial dysfunction and to restore constitutive nitric oxide synthesis.
