Head Dziadevych Serhii V., Доктор біологічних наук Registration Date 20-11-2024 Organization Institute of Molecular Biology and Genetics of NAS of Ukraine popup.description1 The aim of the project is the synthesis, study of structural, physicochemical and catalytic properties of new catalytically active nanomaterials, namely, artificial enzymes (nanozymes); creation on their basis of new diagnostic methods and study of biological action of nanocomposites on eukaryotic cells (in particular, yeast). The creation of new nanomaterials with catalytic properties will allow the development of new methods of highly sensitive quantitative detection of a number of practically important substances (glucose, lactic acid, alcohols) for clinical diagnosis, sports medicine and food technology. popup.description2  A potentiometric biosensor for determination of creatinine content was developed based on the use of commercial and recombinant creatinine deaminase and pH-sensitive field-effect transistors. The main analytical characteristics of the potentiometric biosensor for determining the creatinine content were studied (linear operating range, sensitivity, detection limits, dependence on pH, buffer capacity, ionic strength of the medium, etc.). Optimal compositions of СDI and nanoparticles for improving the sensitivity and stability of biosensor enzymatic systems have been worked out. The obtained composites were characterized by electrochemical methods using a potentiostat/galvanostat. Storage stability was high for all developed immobilization methods and exceeded 100 days of storage. The dependences on the experimental conditions were typical for enzyme biosensors and did not depend on the enzyme immobilization method. It is shown that the linear range of creatinine determination is quite suitable for further work with real samples of blood and dialysis fluid when they are diluted 10 times.
