The primary goal of the project is to develop new biomedical and veterinary materials based on supramolecular systems of the "host-guest" type, rotaxane and pseudorotaxane types, and other topologies using cucurbiturils, cyclodextrins, and their modified analogs as receptors. During the first phase of the project, supramolecular systems based on triazole-containing carboxylic acid derivatives and cucurbiturils with ring sizes of 6 and 7 were synthesized. The formation of complexes was confirmed using thermogravimetric analysis, differential scanning calorimetry, and scanning electron microscopy. The structural features and binding energies of the supramolecular systems were studied using quantum-chemical methods and molecular dynamics. It was demonstrated that, for three cucurbiturils (sizes 6, 7, and 8), the most stable structure involved heterocyclic acids entering the host cavity via their thioacetic fragment. Based on the study of the spectral and photophysical properties of various fluorescent compounds, it was established that the most promising dyes for creating rotaxane systems include acridine orange and its dimers, polymethine, styryl, and triphenylpyrazoline dyes. Squaraine rotaxanes containing both unsubstituted and fluorinated fragments in the macrocycle were synthesized. Encapsulation of the squaraine dye within the macrocycle enhances its photostability and quantum yields. The project involves establishing a new infrastructure unit with the creation of a center for the collective use of scientific instruments, contributing to the further development of supramolecular chemistry in Ukraine.
