In particular, our goal is to investigate the radiosensitizing potential of combinational approaches of arginine-deprivation therapy (ADT) based on the sophisticated 3D spheroid assay platform and cellular models of head and neck squamous cell carcinomas (HNSCC). popup.description2  The goal of the I-DRUG project is to develop new innovative metabolic strategies that improve the therapeutic potential of radiation therapy for cancer, in particular, to study the radio sensitizing potential of enzymatic starvation for the amino acid arginine (ADT) in cell models of highly metastatic human head and neck squamous cell carcinomas (HNSCC). Our working hypothesis is the assumption that the anticancer efficacy of ADT can be improved by the rational design of combined approaches involving the recombinant arginine-degrading enzyme arginase (rhARG1) and certain additional chemotherapeutic factors, in particular plant-derived arginine analogues. Within the framework of the first stage of the project, a two-stage scheme for obtaining a purified rhARG1 drug based on constructed by us yeast producers was optimized, the advantages of which are high stability and efficiency as a component of ADT. Also, a unique cell model was created based on the subline of HNSCC SAS-R9 to study the mechanisms of radio sensitization, which is the main goal of the I-DRUG project. Extracts of the leguminous plant Indigofera spicata containing Arg analogues as components of combined ADT were obtained.
