Di and three functional orthogonally protected building blocks were synthesized for a new direction in the search for lead compounds and DNA-encoded libraries. Original synthetic approaches to new building blocks that can be used for the design of lead-like libraries have been developed, in particular, bicyclic conformationally hindered 3D compounds combining fragments of saturated heterocyclic amines and hydrophilic aromatic heterocycles. Methods are proposed that will allow using these objects in conditions of parallel synthesis with the aim of obtaining lead-like libraries of compounds that are usually characterized by increased hydrophilicity.
