Kukhar Institute of Bioorganic Chemistry and Petrochemistry of the National Academy of Sciences of Ukraine popup.description1 The project is devoted to understanding of the nature of primary cellular reactions to a number of phytohormones, polyamines and the role of phosphatidylinositol 4-kinases (PI4K) and EDS1 (ENHANCED DISEASE SUSCEPTIBILITY 1) proteins in production of cell signaling second messengers during the regulation of plant metabolism. Solving these goals anticipates the investigation of molecular mechanisms that transduce a phytohormonal signals in plant cells inducing a range of consequtive changes in intensity and direction of metabolic pathways leading to plant adaptation. Special attention will be paid to examination of the role of phosphatidylinositol 4-kinases and EDS1 proteins in formation of cell signaling second messengers in response to a range of phytohormones with application of transgenic Arabidopsis thaliana plants with mutation in phosphatidylinositol 4-kinases (PI4K) genes: pi4kβ1β2, sid2pi4kβ1β2, pi4kb1b2sid2eds1. Investigations of the role of phospholipases with changed expression of their isoforms are also planned in regulation of key enzymes of proline metabolism in response to polyamines. Analysis of specific responses of serine-rich peptides SCOOP is envisaged in regulation of cellular metabolism of Arabidopsis thaliana plants. Changes in phospholipid profile in Arabidopsis thaliana will be investigated during plant response to elicitors and hemi-biotrophic fungal infections with the help of genetically-inserted fluorescent markers. Investigations of the role of diacylglycerol kinases with changed expression of their isoforms will be also conducted in regulation of cell metabolism in response to polyamines. popup.description2 The results obtained indicate the key role of biosynthesis (SID2) and signaling genes (EDS1, PAD4) in providing resistance of Arabidopsis plants to the attack of the bacterial pathogen Pseudomonas syringae. The hormone brassinosteroid induces resistance in a transgenic line with the deletion of the salicylic acid (SA) biosynthesis gene SID2. New data on the role of phospholipid signaling, in particular PI-4 kinases, in mediating cross-hormonal interactions between brassinosteroids (BRs) and SCs in the formation of the defense response were obtained. The constitutive deficiency of PI4Kβ1β2 in pi4kβ1β2 mutant plants increased the susceptibility to an actin-degrading drug, and the leading role of salicylic acid in these processes was established using complex mutants sid2pi4kβ1β2, eds1sid2pi4kβ1β2, sid2pi4kβ1β2, and pad4sid2pi4kβ1β2. The activation of expression of genes associated with defense responses, wound stress marker genes, and a number of plant senescence marker genes under the influence of latB was shown, indicating the involvement of the actin cytoskeleton in the cross-regulation between leaf senescence and plant immunity. For the first time, it was found that isozymes of phospholipase D type PLDγ play a role in the realization of polyamine action in Arabidopsis thaliana cells in the processes of formation of resistance to salt stress and production of secondary lipid mediators. Realization of the biological action of SCOOP peptides in suspension cultures of Arabidopsis thaliana cells occurs due to the production of phosphatidic acid and the formation of reactive oxygen species. SCOOP peptides 2, 7, 12 are characterized by the highest level of activation of reactive oxygen species production in suspension cultures of Arabidopsis thaliana cells.
