Registration Date 09-01-2025 Organization Institute of Molecular Biology and Genetics of NAS of Ukraine popup.description1 This project aims to develop fundamental and technological principles for the production of sensor platforms based on graphenized textiles for the analysis of clinically important molecules in human sweat. To achieve this goal, we propose to use unique samples of conductive fractionated graphene flakes as an electrode material for the production of biocompatible amperometric textile electrodes. To make the electrodes sensitive and selective to the target metabolites (glucose and lactate) the developed textile electrodes will be covered with appropriate oxidoreductases. Further on, the performance characteristics of the created textile electrodes will be tested in samples of artificial sweat. popup.description2 Modern diagnostics of clinically important biomarkers shows a tendency to switch from invasive tests to non-invasive flexible platforms. Making of such a platform usually require complex procedures: creating a template, printing electrodes on a flexible matrix and subsequent functionalization of the surface for getting a signal. Within the framework of a project enzyme nanocomposite electrodes were created for the first time on the basis of graphene textile manufactured by Grafren AB. They were used as amperometric biosensors for determination of glucose and lactate. Their advantages in comparison with analogues based on screen-printed carbon and gold electrodes modified with graphene were shown. Modification of the commercial screen-printed carbon electrodes with graphene textile was developed and sensitivity modified electrodes to hydrogen peroxide was determined. The process of immobilization of glucose oxidase and lactate oxidase on the surface of graphene textile was optimized. pH 7.4 was found as optimal operating conditions for determination of glucose and lactate using graphene textile biosensors. They showed high sensitivity – 1.99 µA/mM and 2.82 µA/mM for glucose and lactate respectively and ability to operate in a wide range of pH (5.5–8.0). Operational stability of biosensor for determination of lactate was found as 55 measurements. This biosensor retained 90% of initial activity when being stored in 0.1 M PBS, pH 7.4 at + 40C for a month. Influence of the interfering components of sweat such as: uric acid, urea, ammonium chloride was examined. Additional fabrication of membranes with poly-m-phenylenediamine substantially reduced the influence of the interferents on the measurements. Procedure for determination of lactate content in artificial sweat samples was developed. Additional modification of graphene textile electrodes was studied. Fabrication with gold nanoparticles increased the sensitivity of lactate determination by 6.5 times up to 6.6 µA/mM. Product Description popup.authors Biloivan Olha A. Bubka Kateryna A.
