The goal is to investigate the antitumor mechanisms and immunomodulatory properties of a combined immunotherapy regimen based on PCPs and DCs in an experiment. Research methods are cell culture method, cytofluorimetric, molecular genetic, statistical. Equipment is flow cytofluorimeter Navios EX, Beckman Coulter, USA, centrifuges, microscope, WPS 600/C and VT scales, Nano Drop 1000 spectrophotometer, Varioskan LUX. A methodological approach was developed to obtain a lysate of tumor cells of the MDA-MB-231 line using PCP LMB033. It was shown that the cytotoxic effect of the closed, and to a much greater extent the open form of PCP, on tumor cells is accompanied by the release of DAMPs. Methods for loading DCs with tumor cell lysate obtained under the action of FCP have been developed. It has been established that the closed form of LMB033 at both 16 and 32 µM concentrations contributes to an increase in the level of costimulatory molecules CD86, HLA-DR, the CD83 maturity marker, and the density of HLA-ABC molecules on the DC membrane. It has been established that the use of the open form of FCP LMB033 for the treatment of tumor cells contributed to an increase in the degree of DC maturity, increased expression of costimulatory molecules - HLA-DR and CD86 on their membrane only when FCP was used at a concentration of 16 µM. Loading DCs with a lysate of MDA-MB-231 tumor cell line treated with the LMB033 peptide in its open or closed forms did not statistically significantly affect their viability. It was found that the DC and FCP construct affects both the number and functional activity of the CD3-(16+56+-cells) natural killer cell (NKC) population among autologous lymphocytes after the mixed leukocyte culture (MLC) reaction.
