During the work, the possibility of cultivating MSCs BM on bone implants and directed osteo-differentiation of MSCs BM in the system of monolayer (2D) and spheroid (3D) growth was investigated. The aim of the study was to compare the osteo-differentiation ability of MSCs BM in the systems of monolayer (2D) and spheroid (3D) growth. The objectives of the study were to develop protocols for cultivating MSCs in monolayer (2D) and spheroid (3D) growth systems, to compare the osteodifferentiation efficiency of MSCs in different cultivation systems and in the presence of bone substitutes. To conduct experimental directed differentiation of MSCs in monolayer (2D) and spheroid (3D) growth systems according to the osteogenic phenotype. As a result, a protocol for cultivating MSCs on bone granules was developed and tested to increase the efficiency of osteodifferentiation of cells, and the survival, proliferative potential, and ability for directed osteodifferentiation of MSCs in different cultivation conditions were compared: monolayer, spheroids, in the presence of bone substitutes. The scientific and practical significance of the work lies in the development of protocols for osteodifferentiation of BM MSCs in the presence of bone substitutes and in monolayer and spheroid growth systems, which enables more effective use of osteodifferentiated BM MSCs for bone tissue restoration in medical practice.
