Correlation among the following factors was calculated – genotype, culture medium, root length, sprout length, adaptability to ex vitro conditions, and seed production in spelta. Taking into account all the indicators of in vitro culture, namely, the frequency of regeneration, rooting of regenerants, their adaptation to septic conditions and seed production, the best genotype for biotechnological research of spelta was selected that is the breeding line No. 3865 (out of 5 tested) for the use of combinations of MS/MSBA or N6/MSR (out of 8 tested) culture media for the cultivation of immature embryos. Phylogenetic analysis of spelt and bread wheat samples was performed. For the first time, a phylogenetic tree was constructed that includes not only modern zoned varieties, but also diversified varieties of wild spelt. According to the results of microsatellite analysis at loci Xgwm 219, Xgwm 261, Xgwm 383, Xgwm 469, Xgwm 508, Xgwm 626, the absence of species differences between bread wheat and spelt was shown, and, accordingly, they belong to the same species. The morphology and number of stomata on the abaxial and adaxial sides of the leaf of emmer, spelt and bread wheat of Zymoyarka variety were studied. A significant difference in the number of stomata per unit area of the adaxial surface of the leaf was found in T. aestivum of Zymoyarka variety, T. spelta var. album and T. dicoccum var. aeruginosum. The highest number of stomata per unit area of the adaxial surface of the leaf was observed in spelt, and the lowest - in wheat. On the abaxial surface of the leaf, the number of stomata per unit area was significantly higher in spelt and did not differ in wheat and spelt. Product Description popup.authors Bannykova Mariia O.
