The PS chlorine E6 is incorporated into the cytoplasm of GB cells of the U251 line and cells of HEK293 line (reference cells), the fluorescence intensity is comparable, in contrast to the much weaker fluorescence of tumor-untransformed fetal rat brain cells (E14, used as a control for intracellular incorporation of PS). Direct exposure to chlorine E6 (1, 2 and 3 μg/ml) and direct exposure to LI ( = 660 nm, power 0.4 - 0.6 W, dose (10 - 75) J/cm2, continuous or pulsed mode) dose-dependently increases cytodestructive and antimitotic effects in U251 cell culture. The level of cytodestructive and antimitotic effects is significantly lower in cultures of non-tumor cells of the HEK293 line. Combined exposure to chlorine E6 and LI in different modes dose-dependently decreases the viability and mitotic activity of tumor cells in U251 culture. An effective mode of photodynamic exposure to achieve cytodestructive and antimitotic effects in human GB cell culture of the U251 line is the combined application of an irradiation dose of 25 J/cm2, with a power of 0.6 W in a pulsed mode with preliminary incubation of the cell culture with chlorine E6 at a concentration of 2 μg/ml for 4 h, which is not irreversibly destructive and relatively safe for HEK293 cell cultures (reference cells). For primary human GB cell cultures under combined exposure to chlorine E6 (2 μg/ml) and LI (0.6 W), an effective cytodestructive effect is achieved: in the case of primary diffuse astrocytoma (G3) cells - when using 10 J/cm2 in a pulsed mode; for primary GB (G4), primary gliosarcoma (G4), recurrent astrocytoma (G3) and recurrent oligodendroglioma (G3) cells - 25 J/cm2 in a pulsed mode.
